Participants, who had gone through at least two depressive episodes but were currently in remission, took either prucalopride or a placebo for a duration of seven to ten days. The investigators concentrated on the drug’s influence on memory and cognitive processing, due to its action on the 5-HT4 serotonin receptor present in both the gut and the brain areas integral to memory, such as the hippocampus.

The trial indicated that individuals receiving prucalopride exhibited better performance in tasks assessing word recall, working memory speed, and facial recognition in comparison to those who received the placebo. Unlike conventional antidepressants that influence emotional processing, prucalopride seemed to enhance cognitive abilities associated with attention and memory while leaving emotional processing unchanged. This points to a distinct mechanism, possibly involving elevated levels of BDNF, a protein associated with neuronal health.

In spite of these encouraging findings, researchers warned that the small sample size, brief duration, and specific demographic characteristics limit the applicability of the results. While prucalopride’s influence on persistent cognitive difficulties related to depression could lower the risk of relapse, additional studies are necessary to investigate long-term effects and wider applicability. Should future research validate these advantages, prucalopride might offer a new strategy for addressing cognitive deficits in depression and other related disorders.

The research carried out at the Warneford Hospital examined the possible cognitive advantages of a medication called prucalopride, which is generally prescribed for chronic constipation, for individuals who have experienced a history of depression.
